expression of intercellular adhesion molecule 1 and vascular adhesion molecule 1 in kidney of patients with lupus nephritis and membranoproliferative glomerulonephritis immunohistochemical and computer imaging analysis techniques were used to detect the expression of intercellular adhesion molecule 1 ( ICAM 1 ) and vascular adhesion molecule 1 ( VCAM 1 ) in the kidney of 25 patients with lupus nephritis ( LN ) type IV and in 15 patients with membranoproliferative glomerulonephritis ( MPGN ) type I . results showed that ICAM 1 and VCAM 1 expressions were increased significantly in the kidneys of patients with MPGN and LN and that ICAM 1 in the glomeruli of LN patients correlated well with glomerular endothelial cell proliferation . ICAM 1 and VCAM 1 may well take a role in the pathogenesis of LN and MPGN .

In situ expression of intercellular adhesion molecule 1 in human glomerulonephritis We conducted immunocytochemical analysis with in situ hybridization technique to investigate the presence and role of intercellular adhesion molecule 1 ( ICAM 1 ) in normal and diseased kidneys . A total of 64 renal biopsy specimens were classified in three groups according to the degree of cellular proliferation in glomerulus . group A presented no or mild proliferation ( n 38 ) . group B showed definite proliferation ( n 21 ) . group C had glomerular sclerosis ( n 5 ) . The ratio of cases exhibiting increased glomerular ICAM 1 expression in lupus nephritis ( 9 / 10 ) was significantly different from that in minimally changed disease ( 1 / 5 ) ( P 0 . 017 ) . glomerular ICAM 1 expression level correlated well with the cellular proliferation and infiltration in glomerulus ( B A : P 0 . 001 ; C B : P 0 . 05 ) . The percentage of cases with tubular ICAM 1 positivity in group B ( 81 . 0 ) and group C ( 100 ) was greater than that in group A ( 52 . 6 ) , which was also correlated with that of interstitial cell infiltration . image analysis for intercellular adhesion molecule 1 expression in MRI / lpr mice : effects of chinese herb medicine To clarify the role of intercellular adhesion molecule 1 ( ICAM 1 ) on pathogenesis of autoimmune lupus nephritis in MRL / lpr mice and the effects of chinese herb medicine ( stragalin ) , a computer image analysis system was used to study immunodepositions of ICAM 1 , immunoglobulins and C3 in renal tissue sections of the mice . ICAM 1 was found in the mesangial area and deposited along the glomerular capillary walls in MRL / lpr mice . The distribution intensity of ICAM 1 , immunoglobulins and C3 were significantly decreased after treatment with stragalin in form of decoction per os in the mice . We found that ICAM 1 might play an important role in pathogenesis of autoimmune lupus nephritis in MRL / lpr mice and chinese herb medicine has some inhibition effects on immunodepositions in renal tissue of MRL / lpr mice .

primary culture of proximal tubular cells ( PTC ) from normal mouse kidney as an in vitro model to study mechanisms of development of tubulointerstitial nephritis . induction of ICAM 1 in PTC by antigen primed lymphocytes cultured PTC clearly expressed intercellular adhesion molecule 1 ( ICAM 1 ) under the influence of tubular basement membrane antigen ( TBM ) primed lymphocytes . these TBM primed lymphocytes also demonstrated a high cytotoxic activity against cultured PTC . A pure preparation of isolated PTC from BALB / c mouse kidney was brought into primary culture . PTC was prepared by the method of boogaard PJ et al , and our modification . briefly , kidney was perfused with buffer containing 0 . 08 ( w / v ) collagenase . The cortical tissue was then filtered through nylon gauze . viable PTC were separated from other materials by isopycnic centrifugation on a discontinuous nycodenz gradient . The confluent monolayer of PTC showed a typical epithelial morphology with cobblestone like cells in the center of the cell islands . typical dome formation was observed in PTC cultures . these cells also strongly expressed gamma glutamyl transpeptitase activity . coculture of PTC with syngeneic lymphocytes primed with TBM antigen induced ICAM 1 expression in PTC . The effect of fk506 ( tacrolims hydrate ) , an immunosuppressive agent produced by streptomyces tsukubaensis , on crescentic type anti glomerular basement membrane ( GBM ) nephritis in rats was investigated . When rats were treated with fk506 from 1 or 20 days after the anti GBM serum injection , fk506 inhibited the increase in urinary protein excretion . histological observation demonstrated that fk506 suppressed glomerular alterations . In the fk506 treated rats , antibody production and rat IgG and C3 deposits on the GBM were significantly less than those in the nephritic control group . fk506 treatment suppressed the accumulation of ED 1 positive cells , CD4 positive cells , CD8 positive cells , interleukin 2 ( IL 2 ) receptor positive cells , leukocyte function associated antigen 1 ( LFA 1 ) positive cells and intercellular adhesion molecule 1 ( ICAM 1 ) expression in nephritic glomeruli . however , in the in vitro study , fk506 failed to inhibit the up regulated ICAM 1 expression on endothelial cells in response to tumor necrosis factor ( TNF ) alpha . To determine whether altered expression of ICAM 1 occurs in autoimmune lupus nephritis , we studied ICAM 1 expression in kidneys of normal and autoimmune MRL lpr and ( NZBX NZW ) F1 ( NZB / W ) mice . By immunoperoxidase staining , ICAM 1 is constitutively expressed at low levels in proximal tubules ( PT ) , endothelium and interstitial cells in normal C3H / FeJ mice . In nephritic MRL lpr and NZB / W kidneys , staining for ICAM 1 is increased in the PT , particularly in the brush border , and is prominent in the glomerular mesangium and the endothelium of large vessels . By western blot analysis , ICAM 1 is not detected in the urine of normal BALB / c and C3H / FeJ or autoimmune MRL lpr . In this study , we examined the effects of macrophage colony stimulating factor ( M CSF ) on glomerular macrophages in lipopolysaccharide ( LPS ) induced murine nephritis . Mice injected intraperitoneally with either M CSF plus LPS , LPS alone , M CSF alone or saline every day for 8 days were examined for the degree of urine albumin excretion and lymphocyte function associated antigen 1 positive ( LFA 1 ) cells in peripheral blood as well as renal pathology . From our results , LPS or M CSF combined with LPS emphasized the degree of proteinuria , glomerular deposition of immunoglobulins and mesangial proliferation , associated with accumulation of macrophages in the glomeruli . however , in immunohistological examination of kidneys from these nephritic mice , neither intercellular adhesion molecule 1 ( ICAM 1 ) , which may play an important role in the recruitment of macrophages into glomeruli , M CSF receptor nor the number of LFA 1 cells in peripheral blood was enhanced by M CSF . The tissue distribution of cellular adhesion molecules ( ICAM 1 , ELAM 1 , VCAM 1 ) was studied in specimens from six normal human kidneys and in six biopsies from kidneys with tubulointerstitial nephritis associated with sjögren s syndrome . In addition , the expression of cellular adhesion molecules was examined both in four renal biopsies from cases of tubulointerstitial nephritis of diverse pathogenesis and in six lip biopsies from cases of sjögren s syndrome . ICAM 1 was expressed on vascular endothelial cells in normal kidneys , in all specimens of tubulointerstitial nephritis and in salivary glands . On tubular epithelial cells , ICAM 1 appeared slightly in normal kidneys ; otherwise tubular epithelial ICAM 1 was observed in and around the foci of cellular infiltration in all cases of tubulointerstitial nephritis . The critical role of intercellular adhesion molecule 1 in masugi nephritis in rats . intercellular adhesion molecule 1 ( ICAM 1 , CD54 ) , an adhesion molecule of the immunoglobulin superfamily , is an endothelial cell surface ligand for such leukocyte integrins as lymphocyte function associated molecule 1 ( LFA 1 , cd11a / CD18 ) , Mac 1 ( cd11b / CD18 ) and CD43 . these molecules mediate adhesive interactions between leukocytes and endothelial cells and are critically involved in infiltration of leukocytes into inflammatory lesions . The immunohistochemical expression of ICAM 3 , a recently cloned new member of the immunoglobulin family which also binds leucocyte function antigen 1 ( LFA 1 ) , was examined in 80 needle core biopsies from 35 renal allografts , 7 patients with mesangioproliferative glomerulonephritis , 5 patients with extracapillary glomerulonephritis , 4 patients with interstitial nephritis and 5 patients with diabetic nephropathy and 20 normal kidneys . monocyte response to Th1 stimulation and effector function toward human mesangial cells are not impaired in patients with lupus nephritis . monocytes / macrophages activated by Th1 stimulation such as interferon gamma ( IFN gamma ) and CD40 ligand ( cd40l ) infiltrate the kidney and play a critical role in the progression of lupus nephritis ( LN ) . We examined the monocyte response to Th1 stimulation and their effector function toward activating renal resident cells in patients with LN . following stimulation with IFN gamma granulocyte macrophage colony stimulating factor ( GM CSF ) / recombinant cd40l the production of tumor necrosis factor alpha and IL 12 p70 by PBMC was significantly higher in LN patients . In coculture experiments employing activated monocytes and human mesangial cells , there was a trend toward higher monocyte chemoattractant protein 1 production by lupus monocytes compared to normal controls . basal expression of CD40 , ICAM 1 , and STAT 1 was significantly higher in monocytes from LN patients , suggesting ongoing activation . monocyte response to IFN gamma , as accessed by intercellular adhesion molecule 1 upregulation and phosphorylation of STAT 1 , was comparable between the two groups . Thus , in contrast to earlier reports , Th1 dependent monocyte activation is not impaired . In this disease activated monocytes appear to be fully capable of inducing renal injury .

The antinephritic effects of butein ( 3 , 4 , 2 , 4 tetrahydroxychalcone ) on original type anti glomerular basement membrane antibody associated glomerulonephritis in rats were investigated . butein was given to anti glomerular basement membrane antibody associated glomerulonephritic rats for 15 days after the induction of nephritis . butein prevented proteinuria and histological alterations . The up regulation of intercellular adhesion molecule 1 ( ICAM 1 ) expression and increase in leukocyte function associated antigen 1 ( LFA 1 ) positive cells in nephritic glomeruli significantly declined with butein treatment . In the further investigation to clarify the effects of butein on ICAM 1 expression , human umbilical vein endothelial cells were treated with butein in the presence of tumor necrosis factor alpha ( TNF alpha ) or phorbol 12 myristate 13 acetate ( PMA ) . butein prevented the up regulation of ICAM 1 expression mediated by TNF alpha or PMA on human umbilical vein endothelial cells in a dose dependent manner . glomerular ICAM 1 expression related to circulating TNF alpha in human glomerulonephritis . To clarify the in vivo involvement of cellular adhesion molecules and cytokines in human glomerulonephritis , we have investigated the glomerular and interstitial expression of intercellular adhesion molecule 1 ( ICAM 1 ) in 69 kidney biopsy specimens by immunohistochemical methods and its correlation with serum bioactive tumor necrosis factor alpha ( TNF alpha ) and soluble ICAM 1 ( sicam 1 ) levels in 43 cases . In normal controls , glomerular ICAM 1 expression and serum TNF alpha and sicam 1 levels showed a mean score of 1 . 0 ( n 7 ) and were 12 . 1 / 1 . 5 and 187 / 5 ng / ml ( mean / SEM , n 25 ) , respectively . inflamed site specific delivery of bone marrow derived cells carrying IL 1Ra . A gene delivery system using bone marrow derived cd11b ( ) CD18 ( ) cells and their interaction with adhesion molecules was established . after transplantation into mice , these vehicle cells may be recruited into glomeruli upon lipopolysaccharide ( LPS ) treatment , and the number of recruited cells corresponds to the expression of intercellular adhesion molecule 1 ( ICAM 1 ) in glomeruli . using this system , interleukin 1 receptor antagonist ( IL 1Ra ) was delivered into animal models of glomerulonephritis evoked by anti glomerular basement membrane antibody ( anti GBM nephritis ) . urinary albumin excretion and the serum creatinine level were significantly elevated after anti GBM antibody injection in mock treated mice , whereas they were suppressed in the IL 1Ra treated mice . histological analysis revealed that glomerular injuries were also suppressed in IL 1Ra treated mice . We further confirmed that specificity for inflamed glomeruli was significantly enhanced by a combination of the Cre / loxP system with the IL 1beta promoter . these data suggested that our novel system may be used as a therapeutic intervention for glomerulonephritis .
